Relationship between CRTH2 mRNA expression in peripheral blood and IgE and eosinophil levels in adult patients with asthma

Zusammenfassung

Background: Chemoattractant receptor-homologous molecule expressed on Th2 cells (CRTH2) is a receptor for PGD2 expressed on eosinophils, basophils, macrophages, monocytes, mast cells, Th2 cells, innate lymphoid cells type 2 (ILC2) and dendritic cells. CRTH2 has been involved in asthma, nasal polyposis (NP), and aspirin-exacerbated respiratory disease (AERD). We aimed to investigate the CRTH2 expression in asthmatic patients and to correlate with total IgE and eosinophils. Methods: We included 155 patients with asthma (of them, 37 with NP and 17 with AERD) and 35 non-atopic non-asthmatic controls. All patients signed an informed written consent. Total RNA was isolated from peripheral blood and the CRTH2 gene expression was determined by qPCR using SYBR Green. Values were obtained using the comparative ΔΔCt method with GAPDH as reference gene Results: Asthma patients showed elevated levels of CRTH2 mRNA (8.38±7.52) compared to controls (4.30±2.32) (p<0.001). Asthmatics with NP showed higher expression of CRTH2 (11.37±10.69) (p<0.001). Patients with AERD also showed higher levels of CRTH2 (10.00±12.6) (p=0.029). There were positive correlations between CRTH2 expression and both total serum IgE (0.386) (p<0.001) and eosinophil counts (0.658) (p<0.001) Conclusions: CRTH2 mRNA levels were significantly increased in peripheral blood of patients with asthma, with higher levels in patients that concomitantly had NP. The expression of CRTH2 partially correlated with total IgE and peripheral blood eosinophil counts. CRTH2 expression levels might be a clinically useful biomarker of asthma, NP and AERD and might assist in the selection of specific antagonist therapies
