Age-dependent changes in the expression of Trkb by human Meissner corpuscles

Résumé

Unlike other types of sensory corpuscles and nerveendings, which have very complex neuronal and growth fac-tor dependence, the cutaneous Meissner corpuscles dependexclusively on the NT system TrkB/BDNF/NT-4. However,the pattern of expression of TrkB in human Meissner corpus-cles in not known. The experiments in this study were desig-ned to pursue further the findings that suggest that BDNFand NT4 have critical roles in the development and mainte-nance of Meissner corpuscles, by analyzing the pattern ofexpression of TrkB, their high-affinity receptor, in human gla-brous skin at different ages from 8 weeks of life to old sub-jects. In developing Meissner corpuscles TrkB was expressedin the axon supplying the corpuscles (until 18 months of age).Thereafter there was a switch in the localization of TrkBwhich was restricted to the lamellar cells of Meissner corpus-cles. Importantly that the number of Meissner corpuscles thatexpress TrkB significantly decreases in subjects older than 50years. All together these results lend support to the role ofBDNF and NT4 in mechanoreception
